Syntrophins are modular adapter proteins that function by recruiting signaling molecules to the cytoskeleton via their direct association with proteins of the dystrophin protein family. Each of the five syntrophins (α, β1, β2, γ1, γ2) consists of two pleckstrin homology (PH) domains, a postsynaptic density-95/Discs large/zona occludens (PDZ) domain, and a syntrophin unique (SU) region.
Beta 2 syntrophin plays together with alpha syntrophin a role in forming and maintaining neuro-muscular junctions’ structure and both syntrophin can partially compensate for the loss of the other as shown by the phenotype of the knockout mice. In addition it regulates the exocytosis of secretory granules via its interaction with PTPRN. Beta 2 syntrophin has been suggested to bind and organize the subcellular localization of a variety of membrane proteins including sodium channels, ATP –binding cassette transporters and potassium channels by binding via its PDZ domain to the C-terminal PDZ binding site of these receptors..
