PTPRB: Human tyrosine receptor phosphatase beta (PTPRβ)

PTPRβ belongs to the subtype R3 family of tyrosine receptor PTP’s which has currently 8 members in human. This tyrosine phosphatase family consists of enzymes that contain a large extracellular domain composed of multiple fibronectin type III repeats, a single transmembrane segment and one intracytoplasmic catalytic domain. The mouse homologue of PTPRβ has been shown to interact with the angiopoietin receptor Tie-2 and with VE-cadherin, which is an essential adhesion molecule in endothelial adherens junctions. This interaction is mediated by the membrane-proximal extracellular domains of each protein, in independent of the enzymatic activity of this phosphatase and reduces the tyrosine phosphorylation of VE-cadherin resulting in cell layer permeability. PTPRβ has been suggested to play a potential role as a regulator of blood vessel morphogenesis and maintenance and is a candidate gene for inherited vascular malformations related to the Tie-2 gene.
