PTK9L was identified by its interaction with the catalytic domain of protein kinase C-zeta. It is closely related to twinfilin (PTK9), a conserved actin monomer-binding protein that regulates cytoskeletal dynamics. Like twinfilin PTK9L is composed of two actin-depolymerization factor homology (ADF-H) domains which bind exclusively to G-actin but not like the related ADF/cofilins to both G and F-actin. RT-PCR and Western blot analysis detected PTK9L in all human tissues and cells tested and immunofluorescence revealed spotty accumulation of PTK9L in the cytoplasm close to nuclei.
Here we present the structure of the N-terminal ADF-H domain of PTK9L at 1.7 Å resolution.
